基于高通量多重串联PCR技术的转基因玉米筛选研究

A high-throughput multiplex tandem PCR assay for the screening of genetically modified maize

中文摘要:由于转基因生物越来越受到人们的重视,因而对高通量检测方法的需求越来越大。本文利用一种高通量、低成本的多重串联PCR对14个目标转基因玉米进行了筛选试验,并利用SYBR Green I方法对7个转基因共同筛选要素、6个转基因玉米品种和玉米参考基因进行了定量。研究结果表明,多重串联PCR技术可有效地用于食品和饲料中转基因玉米的筛选。
外文摘要:Genetically modified organisms (GMO) are increasingly gaining acceptance, which has led to an increasing demand for high-throughput methods for their detection. This study describes development of a high-throughput and low-cost multiplex tandem PCR (MT-PCR) assay for the screening of transgenic maize of 14 targets, containing 7 genetically modified (GM) common screening elements, 6 transgenic maize events and a maize reference gene, using SYBR Green I for quantification. The 14-target high-throughput multiplex tandem PCR assay successfully amplified products from single-event samples with a GM maize DNA amount less to 0.001 ng, and displayed high specificity. This assay can efficiently be used for screening GM maize in foods and feeds.
