外文摘要:The aim of this study was to evaluate the effects of RNA-mediated resistance in the transgenic wheat N-12 on rhizosphere microbial communities. Field experiments were performed in two different fields during the entire growing season of winter wheat, and the fields were in a principal wheat yellow mosaic disease area in the eastern region of China. Rhizosphere soils were collected at four developmental stages (i.e., seedling stage, stem elongation stage, flowering stage, and ripening stage), and the rhizosphere microbial communities of various samples were investigated using denaturing gradient gel electrophoresis (DGGE) of 16S rRNA genes followed by DNA sequence analysis and Biolog-EcoPlates (TM) analysis. The General Linear Model (GLM) combined with unweighted pair group method using average linkages (UPGMA) based on the DGGE profiles revealed significant differences based on growing seasons, while no differences in rhizosphere microbial community structures were detected between the crop genotypes. In addition, Canonical correspondence analysis (CCA) results indicated that the rhizosphere bacterial diversity was highly correlated with various environmental factors, including water content, available phosphorus and pH. Principal component analysis (PCA) of the Biolog data demonstrated that seasonal shifts in rhizosphere microbial communities were observed in both treatments. In conclusion, wheat rhizosphere microbial communities were significantly influenced by different factors, such as growing stage, soil physical and chemical properties and field type, and no differences were observed between the transgenic wheat and its parental line.
