中文摘要:本发明公开一种使用改进孟加拉红培养基检测水稻种子携带真菌的方法。该方法用改进孟加拉红培养基培养检测水稻种子,其培养是将培养皿放入培养箱中,25℃,光照强度为9000Lx,光照12h/黑暗12h条件下培养1d,待种子吸涨后,再置于?20℃冰箱中保持12h,取出培养皿在28℃恒温,光照强度为9000Lx,光周期为12h光期/12h暗期,培养6d,挑取真菌,显微镜下观察检查种子携带的真菌种类。所述改进孟加拉红培养基增加了硫酸铜,硝酸锌,尸胺,减少了硫酸镁用量。本发明方法有利于不同种类真菌的生长,可多检测分离到真菌种类,操作不易被细菌污染,真菌菌落直径较小,便于真菌的分离和统计。

外文摘要:NOVELTY - Detecting rice seeds with fungi involves preparing rose Bengal medium by adding the ingredients in the bottle and mixed with 1L water. The rose Bengal medium is sterilized at 121 degrees C, and 0.1-0.15 megapascal. The rose Bengal medium is prepared by adding 5 g peptone, 10 g glucose, 1 g potassium dihydrogen phosphate, 0.25 g magnesium sulfate heptahydrate, 15 g agar, and 15 g granules to a 9 cm diameter dish and mixed with 0.133 g chloramphenicol, 0.1 g chloramphenicol, 0.05 g copper sulfate, 0.029 g zinc nitrate, and 0.0001 g cadaver to obtain the rose Bengal medium.

DETAILED DESCRIPTION - Detecting rice seeds with fungi involves preparing rose Bengal medium by adding the ingredients in the bottle and mixed with 1L water. The rose Bengal medium is sterilized at 121 degrees C, and 0.1-0.15 megapascal. The rose Bengal medium is prepared by adding 5 g peptone, 10 g glucose, 1 g potassium dihydrogen phosphate, 0.25 g magnesium sulfate heptahydrate, 15 g agar, and 15 g granules to a 9 cm diameter dish and mixed with 0.133 g chloramphenicol, 0.1 g chloramphenicol, 0.05 g copper sulfate, 0.029 g zinc nitrate, and 0.0001 g cadaver to obtain the rose Bengal medium. The obtained rose Bengal medium is solidified. The rice seeds of good variety are selected and subjected to surface-disinfectant treatment for 8 minutes using 5 wt.% sodium hypochlorite solution to obtain disinfectant rice seeds. The obtained disinfectant rice seeds are homogeneously placed on a culture medium in a culture dish. The dishes are then placed in a light incubator f! or 1 day at 25 degrees C which involves keeping in light for 12 hours and in dark for next 12 hours to obtain pre-processed rice seeds. The light intensity is controlled to 9000 Lx. The obtained pre- processed rice seeds are placed in a refrigerator at -20 degrees C for 12 hours. The culture dishes are then taken out at 28 degrees C and exposed to photo period of 2 hours light and 12 hours dark period to obtain processed seeds. The intensity of light is controlled to 9000Lx. The processed seeds are cultured in the prepared rose Bengal medium for 6 days, then the fungi are picked, and the morphological characteristics were observed under the microscope to obtain the desired product.
