中文摘要:本研究早期开发了一种幽门螺旋杆菌表位疫苗CUA,该疫苗具有较高的疫苗原性和免疫特异性。为探索CUA疫苗的佐剂效应、提高疫苗的免疫原性,本研究制备了一种壳聚糖纳米颗粒,并分析其作为口服CUA疫苗传递系统的效果。结果表明,壳聚糖纳米颗粒在体外能够显著诱导IFN-γ和IL-17细胞因子的表达,刺激产生抗原特异性IgG、IgA,促进黏膜分泌IgA;同时,壳聚糖纳米颗粒改变了IgG2a/IgG1比值,并参与Th1/Th17-介导的保护性免疫反应。因此,壳聚糖纳米颗粒是一种合适的口服疫苗佐剂,适用于幽门螺旋杆菌表位疫苗。
外文摘要:We previously developed Helicobacter pylori epitope vaccine CUA (CTB-UreA, intra-molecule adjuvant cholera toxin B subunit and epitope UreA((183-203)) from Helicobacter pylori UreA subunits) which possesses high immunogenicity and immunological specificity. In order to
elicit the adjuvantivity and improve the immunogenicity for the CUA vaccine, chitosan nanoparticles were prepared in this current study and used as delivery system for the oral CUA vaccine. Results from this current study indicated that the chitosan nanoparticles significantly induced IFN-gamma and IL-17 cytokines in vitro. The nanoparticles also in vivo induced antigen-specific IgG, IgA and mucosal secretory IgA antibody production. Moreover, the chitosan nanoparticles adjuvant changed the IgG2a/IgG1 ratio and conferred Th1/Th17-mediated protective immune responses. The chitosan nanoparticles may serve as a suitable oral vaccine adjuvant for Helicobacter pylori vaccine.
