外文摘要:A fusion protein is provided which is based on a self-assembling gene- regulatory NSP10 protein and a protein or peptide capable of being fused to NSP10 without interfering with the assembly or aggregation of the resulting fusion protein. The disclosure also relates to any nanoparticle formed thereby whether complete or not, and methods for the use of the NSP10 fusion protein are also disclosed, including use as vaccines for any indication in humans or animals, therapeutic methods involving the use of the fusion proteins such as using the protein to targeted an antibody or receptor, such as for treating or diagnosing cancer, biosensors using the fusion protein, or the use of the fusion proteins in cell sorting or any imaging application.
NOVELTY - A fusion protein comprising a self-assembling non-structural protein 10 (NSP10) and a protein or peptide capable of being fused to NSP10 without interfering with the assembly or aggregation of the resulting fusion protein; or a fusion protein comprising a self-assembling gene-regulatory NSP10 protein and a small therapeutic molecule situated within a hollow hydrophobic core of NSP10, is new.
USE - The protein is useful for preparing vaccine composition, immunogenic composition, or pharmaceutical composition; for enhancing immunogenicity of antigen; for cell sorting; for imaging target material; for enhancing production of recombinant protein of interest; used as an antigen display system for the production of antibodies or the development of vaccines; for displaying antibody or affinity directing proteins or peptides at either or both termini; for carrying internalized imaging agent within its hydrophobic core; used as peptide or protein display systems for applications in biosensors or target directed therapeutics; used as an attachment scaffold where the peptide, small molecule or protein can be attached to the NSP10 protein by a chemical or physical process; used as vaccine and diagnostic agent (all claimed), where vaccine includes anti-parasitic vaccines, anti-insect vaccines, ant-microbial vaccines, anti-protozoan vaccines, cancer vaccines and viral vaccines; and for treating or diagnosing cancer.
ADVANTAGE - The tetrahedral arrangement of the expressed proteins allows for larger fusion partners by reducing the likelihood of steric restrictions created by large fusion proteins caused by the smaller surface area of the nanoparticle. Simultaneous fusions also add the advantage of affinity tags located at the terminus opposite the fusion partner.

(1) a vaccine composition comprising the fusion protein and a pharmaceutical vehicle, carrier, or excipient;
(2) an immunogenic composition comprising an immunogenic amount of the fusion protein and a pharmaceutical vehicle, carrier, or excipient;
(3) a pharmace! utical composition comprising the fusion protein pharmaceutical vehicle, carrier or excipient;
(4) a method of preparing an immunogenic composition comprising fusing to NSP10 at least one immunogenic viral protein or peptide capable of being fused to ferritin without interfering with the assembly or aggregation of the resulting fusion protein;
(5) a method of eliciting an immunogenic reaction in a human or animal comprising administering to the human or animal, an immunologically effective amount of the fusion protein;
(6) an isolated nucleic acid sequence encoding for the fusion protein;
(7) NSP10 protein comprising a self-assembling NSP10 protein and a small therapeutic molecule situated within a hydrophobic core of NSP10; or NSP10 protein fused to a protein or fragment from the Apicoplexan or protozoan family of parasites;
(8) a method of enhancing the immunogenicity of an antigen comprising fusing the antigen to an NSP10 prot! ein, wherein the antigen can fuse to NSP10 without interfering! with th e assembly or aggregation of the protein;
(9) a method of cell sorting comprising introducing the fusion protein into a cell sorting apparatus for a time sufficient to allow the fusion protein to bond with a specific type of cell, and then sorting cells based on the bonding;
(10) a method of imaging a target material comprising introducing the fusion protein having an imaging agent to a medium containing the target material and obtaining imaging of the target based on bonding between the fusion protein and the target; and
(11) a method of enhancing production of recombinant protein of interest comprising co-expressing NSP10 in DNA vaccine so as to enhance the recombinant protein of interest.
